I don't know what effect is being used here - maybe some kind of harmoniser or guitar synth? Try 
messing around at the 12th fret, open E, and 5th fret. I play it like this (after seeing a live acoustic 
performance by the man himself on TV)

There's plenty of room for improvisation. It sounds cool if you do have some effects; I like to use a 
faint delay setting to fill out the sound.
